For a subwoofer it should not make any difference. Many subs can be installed backwards. Thats why most decks have a sub polarity reversing mode. HOwever.... most datasheets for subs that specify an optimum box volume already have accounted for the volume that the driver itself displaces, and that assumes normal mounting. So, if you want to mount it backwards you have to subtract 2x the driver displacemnet from the recommended volume.

Are we talking about which direction the sub is facing in the box, or just in the car? I don't see any advantage to invert mounting a sub in a sealed box, especially a prefab box. Maybe looks? But as for what direction the sub faces in general, I would be pointing the sub at the rear of the vehicle.
