Sub Mounting Position

Does the position of the sub have to be centered in the box?
Basically I want to mount both my subs aprox 2 inches off center, so I have room in the center to mount my amp.

go for it...just make sure you brace the box properly underneath the amp rack if it's on the box. Box flex is a killer for amps.

Also keep in mind that some amps don't cool very well when installed vertically (like on the back of a sub box). Many like only to be mounted with the heat sink facing up. (fan cooled don't matter as much obviously).
